T-cell immunoglobulin and ITIM domain receptor (TIGIT) is a surface protein and an inhibitory receptor that suppresses T cell activation by promoting the generation of mature immunoregulatory dendritic cells []. It contains an immunoglobulin tail tyrosine (ITT)-like phosphorylation motif and an ITIM (immunoreceptor tyrosine-based inhibition motif) in its cytoplasmic tail. The identified ligands of TIGIT include the poliovirus receptor (PVR or CD155) and PVRL2 (Nectin2 or CD112). The TIGIT/PVR signaling has been shown to suppresses granule polarization and NK cell cytotoxicity [].
