Description: Sestrins (Sesns) are involved in ROS (reactive oxygen species) regulation. In mammals, there are three Sestrins (Sesn1, Sesn2, and Sesn3), while in Drosophila melanogaster and C. elegans, there is only one Sesn []. The N-terminal of Sesns shares protein sequence and structural similarity with the C-terminal α-helical domain of the AhpD protein from Mycobacterium tuberculosis. Interestingly, AhpD is a component of alkyl-hydroperoxide reductase participating in defense against ROS and ROS produced by host immune cells []. Sesn1 (also known as p53-activated gene 26, PA26) was first identified as a target of the p53 protein through a screen []. Subsequently, Sesn2 (also known as hypoxia-inducible gene 95, Hi95) was identified by microarray-based analysis of novel genes activated by prolonged hypoxia []. Both the Sesn1 and Sesn2 genes can be activated by genotoxic stress in a p53-dependent manner []. All three mammalian Sesns can be activated by the transcription factor forkhead transcription factor (FoxO3A) and FoxO1, members of the FoxO gene family [].
