This recessive mutant causes mice under 10 days of age to have flaky tails, often with constrictions which result in amputation of the tail, and external ears which are slightly smaller than normal. The earliest recognizable sign appears at about 2 days after birth when the skin of the feet looks stretched. By 4 weeks of age, if the tail has not been amputated, the mice look perfectly normal except for slightly smaller ears. Histological sections of the skin between 1 day and 2 weeks of age show that the stratum granulosum is thinner than normal. The stratum corneum appears normal, but small differences in this layer might be hard to detect because it flakes off so irregularly. A possible explanation of the gross effects of the mutant is that the stratum granulosum, or possibly the whole epidermis, does not grow quite fast enough to accommodate the rapidly growing young mouse. While this produces no lasting effect on most of the body, in the ears and tail, which have relatively large surface areas, permanent changes result. The mutant is easily classifiable and has good penetrance in the stock in which it is maintained but may have reduced penetrance in outcrosses. Segregation data from the first backcross into strain C57BL/6J shows 167 flaky tails to 205 normals.
