| molecularNote | The endogenous gene was first replaced with an HPRT minigene flanked by loxH and lox511 and then the HPRT minigene was replaced with the human RHO gene fused to GFP via cre mediated recombination. This resulted in replacement of the endogenous gene by human RHO fused to GFP flanked by loxH and lox511 sites. Western blot analysis indicated that expression of the human gene was about 16% that of the endogenous gene. GFP expression is seen in the eyes. |
