description | Transgenic mice on a C57BL/6J background display no obvious phenotype. The protein product is localized to the cell nucleus. Expression is present in the liver of transgenic mice as a result of the presence of the SV40 large T-antigen nuclear localization signal peptide. Hepatocytes stain blue with X-gal staining. Expression can be induced up to 10-fold by administration of heavy metal ions. |
molecularNote | The mouse Mt1 (metallothionein 1) promoter and flanking DNA (-588 to +68) were fused to the bacterial lacZ gene with a nuclear localization sequence. The construct was inserted into the EcoRI site of a vector that carries 10kb of sequence flanking Mt2 and 7kb flanking the Mt1 gene. These flanking sequences contain ribonuclease-hypersensitive sites thought to be important for enhanced expression. |
