| molecularNote | This allele is derived from Apoetm1Khw by mating mutant animals to a cre deleter strain. Resulting progeny are removed of the floxed neo cassette between exons 3 and 4 leaving behind a threonine to arginine substitution at amino acid 79 (p.T79R). This mutation allows the mouse protein to mimic human APOE4 domain interaction. Removal of the neo cassette resulted in normal levels of gene expression comparable to that of wild-type. Expression of the mutant allele was confirmed by Northern blot analysis of several tissue types. |
