molecularNote | This targeted mutation deletes a 6-kilobase genomic DNA segment containing hs3b (Rr155) and hs4 (Rr156), the two most distant of four 3' transcriptional enhancers residing within the approximately 26 kilobase locus control region downstream of the Igh complex (3'RR or Rr152). The original targeting event replaced hs3b and hsb4 with a loxP-flanked neomycin resistance gene cassette. Crossing of chimeric founder mice with mice expressing Cre recombinase under control of the EIIa promoter yielded progeny in which the neo cassette was deleted in the germline. |
