molecularNote | Transgenic mice overexpress a constitutively active form of PKB (gag-PKB (Akt1) in T lymphocytes under control of the human CD2 promoter and locus control region (LCR). The gag-PKB-encoded protein is equivalent to the oncogenic form of PKB, v-akt, and consists of three parts: an in frame fusion of the viral gag protein, a 21-amino acid peptide linker derived primarily from the 5'untranslated region of the endogenous Akt1 gene locus, and Akt1. An NH2-terminal glycine residue of the gag portion of the chimeric protein serves as a site for myristylation. This myristyl group preferentially targets the gag-PKB fusion protein to the cell membrane where it becomes constitutively activated by upstream kinases. Two individual founder lines were established, 5-4 and 4-1. |
