Enables NAD(P)H dehydrogenase (quinone) activity and NADH:ubiquinone reductase (non-electrogenic) activity. Involved in several processes, including protein polyubiquitination; response to lipopolysaccharide; and vitamin K metabolic process. Acts upstream of or within response to oxidative stress. Is active in cytoplasm and nucleus. Is expressed in heart; liver; main olfactory bulb; olfactory epithelium; and skin. Human ortholog(s) of this gene implicated in several diseases, including hematologic cancer (multiple); hypertension; lung disease (multiple); macular degeneration; and small cell carcinoma. Orthologous to human NQO1 (NAD(P)H quinone dehydrogenase 1).
PHENOTYPE: Targeted null mice display increased toxicity to menadione, insulin resistance, an altered intracellular redox status, as well as decreased pyridine nucleotide synthesis, gluconeogenesis and fatty acid metabolism, leading to reduced quantities of abdominal adipose tissue.
