abstractText  Full text of Abstract: 29. A systemic regulatory locus associated with two new structural alleles of murine Beta-glucuronidase in M. musculus castaneus. K. PFISTER1, V. CHAPMAN2 and K. In two congenic strains, constructed by transferring the structural regulatory gene complex [Gus] of Beta-glucuronidase from M. musculus castaneus onto the C57BL/6 inbred strain background, the structure and the regulation of the expression of Beta- glucuronidase were characterized. Two novel structural forms of Beta-glucuronidase were identified, differing in thermolability and, in one case, in antigenicity from any previously known Beta-glucuronidase allelozyme. They were found to have the same molecular activity as the Beta-glucuronidase from reference strain C57BL/6. The results indicate the presence of new alleles of the structural gene of Beta-glucuronidase. Both mutant structural genes in the congenic strains were similarly regulated by temporal and regulatory gene(s). The development course of the Beta-glucuronidase activity expression paralleled that of reference strain C57BL/6 but remained at approx. half the activity level in all organs studied. In kidney, the androgen-inducible rate of synthesis of Beta-glucuronidase was lowered to approx. half of that in C57BL/6. Thus, the two [Gus] haplotypes of the congenic strains appear to carry mutations in a previously unidentified systemic regulatory locus that appears to alter the rate of Beta-glucuronidase synthesis by a constant factor. It exhibits additive inheritance with F1 Heterozygotes showing intermediate enzyme levels.
