molecularNote | A targeting vector was obtained from the European Conditional Mouse Mutagenesis Program (EUCOMM) project. The targeting vector consisted of two homology arms, the 5 arm comprising exon 3 and the 3 arm comprising exons 5 to 8. Exon 4 was flanked by two loxP sequences, and its removal from the genome generates a shift in the open reading frame after splicing and an early termination of translation. The promoterless selection cassette present in the EUCOMM vector was replaced by two elements present in the pACN vector: a neomycin resistance gene under the control of a PGK (phosphoglycerate kinase) promoter and a self-excising Cre recombinase cassette under the control of a testis-specific angiotensin-converting enzyme (tACE) promoter. Because both elements are flanked by loxP sites, this construct enabled the cassette and exon 4 to be deleted in the testis of the male chimeras, producing heterozygous p84+/ germline cells. |
