molecularNote | A targeting vector was designed to insert the sequence encoding cre/ERT (CreER), an IRES-TVA (avian leukosis virus receptor) element, a floxed tk-NEO selection cassette, and an IRES sequence into the ATG start codon of the Myod1 gene. The construct was electroporated into "129SvJ" derived F12 embryonic stem (ES) cells, which were transiently transfected with a Cre recombinase vector to remove the selection cassette. ES cells that had successfully undergone cre-mediated recombination and no longer retained the cassette were injected into C57BL/6 blastocysts. The resulting chimeric animals were crossed to C57BL/6 mice. |
