molecularNote | The transgene is designed with (from 5' to 3') a ~5.5 kbp mouse Myh6 minimal promoter sequence (including the 3' most exon of betaMHC, the first three exons of alphaMHC [up to the start codon] as well as intervening sequences), a CatCh-IRES-lacZ-hGHpA construct (CatCh, internal ribosome entry site, beta-galactosidase and a human growth hormone polyadenylation signal). The calcium translocating channelrhodopsin CatCh [ChR2(L132C)] cDNA sequence used encodes the first 309 amino acids of green alga Chlamydomonas reinhardtii channelrhodopsin-2 (ChR2 or cop4) that has been modified with an L132C amino acid substitution (CTC-to-TGC) designed to cause enhanced Ca2+ permeability. Founder line B5-7 was identified with high sensor expression in cardiomyocytes. |
