Allele : Akap9<tm1b(KOMP)Wtsi> A kinase anchor protein 9; targeted mutation 1b, Wellcome Trust Sanger Institute

Primary Identifier  MGI:5516002 Allele Type  Targeted
Attribute String  Null/knockout, Reporter Gene  Akap9
Transmission  Germline Strain of Origin  C57BL/6N
Is Recombinase  false Is Wild Type  false
Project Collection  KOMP-CSD
description  Cell line EPD0033_5_D02 was successfully used to make chimeric mice. Germline transmission was accomplished. J:198499
molecularNote  The L1L2_gt0 cassette was inserted at position 4018123 of Chromosome 5 upstream of the critical exon 8 (Build GRCm39). The cassette is composed of an FRT flanked lacZ/neomycin sequence followed by a loxP site. An additional loxP site is inserted downstream of the targeted exon 8 at position 4019078. The critical exon 8 is thus flanked by loxP sites. A "conditional ready" (floxed) allele can be created by flp recombinase expression in mice carrying this allele. Subsequent cre expression results in a knockout mouse. If cre expression occurs without flp expression, a reporter knockout mouse will be created. Further information on targeting strategies used for this and other IKMC alleles can be found at http://www.informatics.jax.org/mgihome/nomen/IKMC_schematics.shtml. Cre-mediated recombination removed exon 8 and left the reporter construct intact. Western blot analysis confirmed the absence of protein expression in the pancreas and testis.
