They are the substrate recognition component of the E3 ubiquitin-protein ligase complex. Human VPRBP is part of the CUL4A-RBX1-DDB1-DCAF1/VPRBP complex that mediates ubiquitination and proteasome-dependent degradation of proteins []. VPRBP also has kinase activity and is capable of phosphorylating histone H2A on threonine 120 (H2AT120p) in a nucleosomal context [].
