ASH1/ASH1L are the members of the Trithorax family.In Drosophila melanogaster, ASH1 (absent, small, or homeotic 1) is an epigenetic transcriptional regulator of HOX genes. Drosophila ASH1 has been shown to methylate specific lysines in histones H3 and H4. Mammalian ASH1 has been shown to methylate histone H3[, ]. ASH1L methylates Lys36 of histone H3 independently of transcriptional elongation to promote the establishment of Hox gene expression by counteracting Polycomb silencing []. It can suppress interleukin-6 (IL-6), and tumour necrosis factor (TNF) production in Toll-like receptor (TLR)-triggered macrophages, and inflammatory autoimmune diseases by inducing the ubiquitin-editing enzyme A20 [].
