Title: Ufd1 exhibits the AAA-ATPase fold with two distinct ubiquitin interaction sites.
Volume: 13
Issue: 7
Pages: 995-1005
Interaction Experiment
Description: A complex of mammalian ufd1 and npl4 links the AAA-ATPase, p97, to ubiquitin and nuclear transport pathways.
Protein Domain
Type: Homologous_superfamily
Description: This entry includes Ubiquitin fusion degradation protein Ufd1 from fungi and Ufd1-like proteins from animals and plants. Ufd1 is part of the Ufd1-Npl4 complex that functions as the substrate-recruiting cofactor for Cdc48 segregase. The Cdc48-Ufd1-Npl4 complex is involved in degradation of misfolded ER proteins []. The Ufd1-Npl4 complex has been found to recruit Cdc48 to ubiquitylated CMG (Cdc45-MCM-GINS) helicase at the end of chromosome replication, thereby driving the disassembly reaction [].In humans, Npl4-Ufd1 acts as a cofactor in reducing antiviral innate immune responses by facilitating proteasomal degradation of RIG-I (a viral RNA sensor) []. The Ufd1 N-terminal fragment is composed of two readily identifiable subdomains designated as Nn and Nc subdomains. The Nn subdomain adapts a double-psi beta barrel fold, and the Nc subdomain has a mixed alpha/beta roll structure []. This superfamily represents the Nn domain found in Ufd1 proteins.

Protein Domain
Type: Family
Description: This entry includes phosphatidylinositol 4-kinase gamma 1-8 (PI4KG1-8) from Arabidopsis and related proteins. They are protein kinases that contain the PI3K/PI4K domain. AtPI4Kgamma4 (also known as PI4KG4) has been shown to interact with and phosphorylate UFD1 (ubiquitin fusion degradation 1) and RPN10 (regulatory particle non-ATPase 10) []. In rice PI4KG4 (also known as OsUbDKgamma4) is involved in the control of flowering under long day conditions by promoting degradation of FTIP1 [].This entry also includes uncharacterised bacterial proteins.
