abstractText  Mice homozygous for the Pitx2<egl1> R115L point substitution on this C57BL/6J background provide a fully penetrant model for early-onset glaucoma, developing elevated intraocular pressure by five weeks of age, which subsequently becomes more severe. In order to better visualize the optic neuron progressive loss in this model (Figure 1), we introduced yellow-YFP transgene to this mouse model by continuously backcrossing Tg(Thy1-YFP)16Jrs from strain B6.Cg-Tg(Thy1-YFP)16Jrs/J (JAX Stock No. 003709) for 15 generations. This transgene drives high level of expression of yellow fluorescent protein in motor and sensory neurons and in subsets of central neurons and this new combined strain, B6.Cg-Pitx<egl1> Tg(Thy-YFP)16Jrs/Boc (JAX Stock No. 013544), allows visualization of retinal nerve and ganglion cell degradation by fundus assessment in live mice. Figure 1. Representative images: A) B6.Cg-Pitx<egl1> Tg(Thy-YFP)16Jrs/Boc with optic ring and more fluorescent protein expressed in optic neurons in the fundus and optic cupping in OCT at 2 months of age and B) B6.Cg-Pitx<egl1> Tg(Thy-YFP)16Jrs/Boc with optic ring and less fluorescent protein expressed in optic neurons and optic cupping in OCT at 4 months of age using an in vivo bright field retinal imaging microscope equipped with image-guided OCT capabilities (Micron III; Phoenix Laboratories, Inc., Pleasanton, CA, USA). The green lines are the plane in which the B-scan to the left of the fundus images were taken in the same animal.
