molecularNote | The allele have an IRES enhancer sequence and a CreER(T2) fusion gene inserted immediately downstream of the insulin-like growth factor binding protein 2 locus (Igfbp2) on chromosome 1. The targeting vector contained, from 5' to 3', an IRES enhancer region, a CreER(T2) fusion gene (Cre-ER(T2); Cre recombinase fused to a G400V/M543A/L544A triple mutation of the human estrogen receptor ligand binding domain), and an SV40 polyadenylation sequence. The plasmid was targeted to the 3' UTR of the endogenous mouse Igfbp2 gene via CRISPR/Cas9 mediated gene targeting in C57BL/6N-derived zygotes. Offspring were bred with FVB mice for germline transmission of the allele. The resulting Igfbp2-CreER colony was then backcrossed to FVB mice for at least 6 generations prior to sending males. |
