| molecularNote | This conditional allele was created using the FLEx switch system. A cassette containing the following elements was placed downstream of the wildtype exon 2 (the first coding exon) into intron 2: a lox511 site in forward orientation; a copy of exon 2, with a mutation changing lysine codon 35 to methionine (p.K35M), in reverse orientation; a loxP site in reverse orientation; and a lox511 site in reverse orientation. Another loxP site, in forward orientation, was inserted upstream of the wildtype exon into intron 1. Upon cre-mediated recombination, the wildtype exon will be removed and the mutated exon will be inserted in the forward orientation, taking its place, leaving a loxP site in forward orientation upstream and a lox511 site in reverse orientation downstream. The mutation in the exon renders the encoded peptide kinase-dead. |
