They are characterised by a central extended spectrin-repeat (SR) and a C-terminal Klarsicht/ANC-1/Syne homology (KASH) domain that can associate with Sad1p/UNC-84 (SUN)-domain proteins of the inner nuclear membrane within the periplasmic space of the nuclear envelope (NE) []. Nesprin-1 and nesprin-2 also contain an N-terminal calponin homology (CH) domain that links to filamentous actin (F-actin) []. Full length Nesprin-1 and nesprin-2 are large multidomain proteins. Smaller N-terminally truncated isoforms lacking the CH domains localise to the inner nuclear membrane, while isoforms that lack the KASH domain have been shown to localise to other regions of cells, including focal adhesions [].This entry represents nesprin-2 from animals, including insects and vertebrates. Nesprin-1 and nesprin-2 are key components of the linker of the nucleoskeleton and cytoskeleton (LINC) complex, which plays critical roles in nuclear positioning, cell polarisation and cellular stiffness []. They also regulate endothelial cell shape and migration []. Nesprin-2 interacts with meckelin and mediates ciliogenesis via remodelling of the actin cytoskeleton [].
