Inbred strains differ markedly in sensitivity to cadmium-induced necrosis of the testis. Resistance is determined by homozygosity for a single recessive gene (cdm/cdm). Homozygotes show undamaged testes after a single subcutaneous dose of CdCl2, whereas cdm/+ and +/+ mice show testes with hemorrhagic and necrotic areas. cdm is found in strains A/J, BALB/c, C3H/He, and C57BL/6; most other strains are wild-type (J:5362). The level of cadmium in testes of DBA/2 (+/+) mice 2 hours after treatment is twice as high as in C3H and BALB/c (cdm/cdm) mice. Paradoxically, the cdm/cdm resistant strains are more susceptible to the lethal effects of cadmium treatment than the +/+ susceptible strains (J:31034) and also more susceptible to the teratogenic effects of cadmium (J:6189). Induction by cadmium of metallothionein (MT) in liver of the +/+ DBA/2 strain is higher than in the cdm/cdm C3H and C57BL/6 strains. The low level in C3H appears to be due to increased rate of degradation of MT (J:7122). The difference between DBA/2 and C57BL/6 in cadmium-induced MT level is controlled by a single locus with codominant alleles that shows close linkage with cdm and may be the same locus (J:7123).
