molecularNote  A loxP site, a copy of the wild type genomic sequence containing exons 3-11 including the stop codon and poly(A) signal sequences, an FRT site flanked neomycin resistance gene cassette and a second loxP site were inserted into intron 2. Codons 79P-120R in endogenous exons 3 and 4 were deleted and IRES sequences and the EGFP reporter gene were inserted into the 3' UTR between stop codon and poly(A) signal. This allele expresses the wild type peptide. Only after Cre-mediated deletion of wild type exons 3-11 will it express the mutated peptide and the EGFP reporter.
