MRC Molecular Neurobiology Unit, MRC Centre, Cambridge, CB2 2QH, England The human GABA(A) receptor is the major inhibitory neurotransmitter receptor of the vertebrate brain. Two isoforms of the alpha subunit and one beta subunit are autosomal in the human. We have mapped the third isoform of the alpha subunit, Gabra3, to the proximal mouse X chromosome by pedigree breakpoint analysis in a mouse interspecies backcross, in relation to seven other gene markers in 58 recombinant animals. We have found that Gabra3 maps proximal to Cf-8 and Gdx and distal to DXPas6. Further, in all recombinants between DXPas6 and Rsvp the genotype is identical to that at Rsvp. Our data suggest the order: Ccn - Hprt - DxPas6 - Gabra3/Rsvp - Gdx/Cf-8 - mdx - tel In addition, they predict that the GABA(A) alpha-3 subunit gene is closely linked to the colour blindness locus in man. Linkage analysis has placed X-linked manic depression close to RCP and G6PD in Xq28 and we propose, therefore that it may be a candidate gene for manic depression. |
