molecularNote | A 2.5-kb promoter region from the mouse tyrosinase (Tyr) gene was used to drive expression of the activated human HRAS gene (T24 Ha-ras), containing a single amino acid substitution of a glycine to valine at codon 12, in pigment-producing cells. Two independent founder mice carrying the transgene survived and showed the same distinct phenotype. One of these founders, a male, showed a single or low copy number integration event. The offspring of this male founder were developed into a line that stably expressed the same phenotype. RT-PCR analysis revealed transgene expression in adult skin, brain and spleen tissue.
