A mutation a t the dreher locus, (dr <2J >) We identified an autosomal recessive mutation on the AKR/J background that causes several pleiotropic effects including deafness; a hyperactive neurological behavior characterized by circling, head tossing, and disorientation; and variable tail length. We also noted that the F2 homozygous mutant mice, produced in a linkage cross (see below) involving the C57BL/6J inbred strain, have a belly spot. Finally, the mutation alters Mullerian duct development in females. Gross inspection of sexually mature mutant females revealed underdeveloped Fallopian tubes and uterus - not even morphologically equivalent to normal virgin females. A cursory histological inspection of the Mullerian duct showed hypoplastic development suggesting that Mullerian duct differentiation in mutant females is either incomplete or not maintained. We tested this mutation with the morphologically similar mutation Sst<J> (shaker short-tail), which had been previously allelism tested by Hope 0. Sweet, with the mutation sst described by Wahlsten, Lyons, and Zagaja (J. Hered. 74:421-425, 1983). We found that our mutation and sst<J> are allelic and gave the AKR-derived mutation the gene symbol sst<2J>. Electrophoretic differences between the C57BL/6.J and AKR/J inbred strains have enabled us to map sst to Chromosome 1. The linkage cross consisted of intercrossing Fl mice and determining the genotype for various isozyme loci on the F2 mutant animals. Out of a total of 34 mutant F2 mice analyzed (equivalent to 68 chromosomes obtained in a standard backcross), 13 were PEP-1B, AKP-1B (like AKR); 12 were PEP-lAB, AKP-1B; 8 were PEP-1B, AKP-lAB; and 1 was PEP-lAB,AKP-1AB. Assuming that the one mouse classified as PEP-lAB,AKP-lAB resulted from obtaining a single recombinant chromosome from each parent, the gene order, with percent recombination, is: Pep-1 - 19.1 +/- 4.8 - sst<2J> - Akp-1 - 13.2 +/- 4.1. The Chromosome 1 location of sst<2J> and the morphological features observed in mutant mice suggested that sst<2J> might be allelic with dr (dreher) . We recently confirmed the suspected allelism using +/dr mice kindly provided to us by Dr. G. Truslove, University College, London. We suggest that the sst<2J> mutation be named dreher-2J (gene symbol dr ). (L. Washburn and Eicher) |
