The eye lens obsolescence (Elo) mutation produces a micro- or anophthalmic phenotype and maps very close to Len-1, a locus encoding a gamma-crystallin electrophoretic polymorphism, on chromosome 1. Multiple independent restriction enzyme analyses of DNA from C3H.Elo mice revealed that the pattern of restriction fragments that hybridize to gamma-crystallin is different from the pattern observed in DNA from the congenic partner strain, C3H/HeJ. Furthermore, the RFLPs observed in C3H.Elo appear to be unique to the mutant strain, since we have failed to observe a similar pattern among DNAs from more than sixty inbred strains of mice. This observation suggests that the Elo mutation arose as a deletion or rearrangement event within the gamma-crystallin gene family. (L. Skow and M. Donner, NIEHS) |
