| description | One of three alleles produced using a targeting construct capable of inducing different allelic variants when mated to mice that express cre, flp, or both. Referred to in the literature as Fgf8neo, the presence of the neo cassette in the intron cause a reduction in the amount of Fgf8 expression and is thus considered a hypomorphic allele. Heterozygotes are normal. Homozygotes die within one day of birth. Gastrulation and limb development appear normal, but aspects of brain development are perturb. |
| molecularNote | Insertion of an frt-flanked neomycin cassette into intron 1 and loxP sites flanking exons 2 and 3. In homozygous embryos, both wild-type and aberrantly spliced mRNAs are produced. Less than 45% of the normal levels of wild-type mRNA is produced from this allele. This allele behaves as a hypomorphic allele. |
