abstractText  Full text of Mouse Genome contribution: Research News: 1. Scruffy, Scr, a new mutation on distal Chr 6. Scruffy, symbol Scr, a dominant mutation affecting the coat, originated in a specific locus experiment in which (C3H/HeH x 101 /H) F1 females were X-irradiated with 6 Gy and mated immediately to tester stock males homozygous for the linked gene pairs alpha bp, d se and ru ep. Heterozygotes have a pronounced shiny and unkempt wavy coat first visible from around 12 days. The vibrissae are normal. Penetrance on outcrossing is complete and viability is normal. Scr/+ mice weigh about 25% less than their normal sibs at weaning although birth weights are similar. Red cell counts and haemoglobin values are also similar. Internal examination showed no obvious differences in viscera between Scr/+ and +/+ although the thymus seemed smaller in the former. Homozygotes are also small but have normal viability and fertility and are sometimes distinguishable from heterozygotes by having a slightly wavier coat and slightly curly whiskers. Crosses to the similar mutant matted (ma) showed that they were not allelic. Scr has now been located on Chr 6, linked to lurcher (Lc) and white (Miwh) but showing independent segregation with sightless (Sig). Outcrosses of Lc Miwh +/+ + Scr males and females to wild type gave the following progeny: Scr 68, Lc Miwh 55, Lc Scr 6, Miwh 7, + 14, Lc Miwh Scr 14, Lc 0, Miwh Scr 2, total 166. The latter two classes are presumed to be double recombinants and suggest the order is Lc-Miwh-Scr with an RF of 18.1 +/- 3.0% between Miwh and Scr. The RF of 9.0 +/- 2.2% between Lc and Miwh is in reasonable agreement with the value of 12 cM shown on the locus map of the mouse (Hillyard et al, Mouse Genome 90:8-21). Its distal position on 6 puts Scr close to motheaten (me) a lethal mutant (though with a more viable allele) suffering fr6m an auto-immune disease which leads to an effect on the coat which is rather similar to that of Scr (Green and Shultz, J. Hered. 66:250-258, 1975). Thus it is possible that is an allele of me with a much less severe effect (Beechey and Searle).
