| description | Loss of Ptgir was shown functionally: e.g., a prostacyclin analogue (cicaprost) was used to inhibit platelet aggregation induced by ADP (platelet aggregation was inhibited in wild-type mice, but not in homozygous mutant mice), relaxation of smooth muscle in aortic rings of wild-type and homozygous mutant mice was measured after stimulation with a prostacyclin analogue (cicaprost, again, homozygous mutant mice showed no relaxation upon stimulation). Specificity of the knock-out was demonstrated by functional assay using other prostaglandins (e.g., PGE2). No differences between wild-type and homozygous mutant deficient mice were seen in response to PGE2. |
