The insertion results in a defined duplication of a large region of sequence within the gene. The neo cassette is located between the redundant segments, disrupting expression. Northern analysis of total heart and kidney RNA demonstrated that no detectable transcript was present in homozygous mice. Western analysis from crude cytosolic brain preparations failed to detect protein in homozygous mice. |
