| molecularNote | This allele was generated by breeding mice carrying Ktn1tm1Kpf with transgenic mice expressing cre recombinase in early development. Sequence intervening the floxed neomycin selection cassette inserted into intron 2 and the loxP site in intron 5 (exons 3 through 5) was excised. An aberrant transcript in which exon 2 splices to exon 6 and causes a frameshift mutation that introduces a stop codon was identified via RT-PCR and sequence analysis. Transcript was undetected by Northern blot analysis, putatively due to nonsense-mediated decay (NMD). Western blot analysis confirmed the absence of protein in homozygous mutant mice. |
