| molecularNote | Exon 1 was flanked by an upstream floxed neo cassette and a loxP site inserted into intron 1. Transient transfection of ES cells with cre recombinase expression plasmids resulted in the excision of the neo cassette and exon 1. Flow cytometric analysis of platelets obtained from homozygous mutant mice showed an absence of surface expression. Western blot analysis of protein lysates from platelets and mammary glands confirmed the lack of protien. Neither full-length nor truncated transcript was detected by RT-PCR analysis of mammary gland lysates. |
