| molecularNote | The human APOE isoform 4 cDNA and a floxed neo cassette were knocked into the mouse Apoe locus via homologous recombination. The knock in event deleted part of exon 2, all of exon 3, and part of exon 4 of the mouse Apoe sequence. Expression of the human gene was confirmed by cDNA sequence analysis of liver mRNA and Western blot analysis of serum proteins from mutant animals. Immunohistochemistry of brain sections showed that the human gene was expressed in astrocytes and neutropil of the hippocampus, and in the neocortex. |
