abstractText  The retinal neovascularization 5 mutation (rnv5) was identified in an F2 intercross population from C57BL/6NJ (JAX Stock No. 005304) and C3A.BLiA-Pde6b+/J (JAX Stock No. 001912). C57BL/6NJ is homozygous for the Crb1rd8 mutation. It was found that the Crb1rd8 homozygotes in that population had two distinct phenotypes: those with small retinal spots and those with significant retinal neovascularization by 4 weeks of age, which was called rnv5. After identifying the rnv5 phenotype in the F2 intercross generation, the mutation was maintained by breeding the siblings with the best rnv5 phenotype at each generation until the F5 generation, at which point rnv5 was backcrossed onto C57BL/6NJ. The N1F1 generation mice showed a very good rnv5 phenotype (Figure 1 Left) at 4 weeks of age compared with the C57BL/6NJ control (Figure 1, Right), so we continued to backcross to C57BL/6NJ until generation N6 and then sibling mated to homozygosity for both Crb1rd8 and rnv5. It was found that rnv5 could be transferred by continuous backcross to C57BL/6NJ but when outcrossed to a strain not homozygous for Crb1rd8 the phenotype was lost, proving this a dominant modifier of Crb1rd8 that lacks ocular phenotype in the absence of Crb1rd8 homozygosity. In 2021 sperm was cryopreserved from homozygous males at generation N6F15. This strain, B6N.Cg-rnv5/Boc, is Jax Stock No. 036993.
