| molecularNote | A neo cassette immediately followed by a 2.3 kb albumin promoter/enhancer was inserted downstream of the endogenous somatic promoter and upstream of exon 1 by homologous recombination. The targeting construct was designed such that transcription of the somatic isoform would be controlled by the inserted albumin promoter, which drives expression in the liver. Protein was undetected in all but two of the somatic tissues tested. There was 87-fold increase in the hepatic protein level whereas the renal protein level was approximately 14% that of wild-type mice. The expression of the testes isoform was unaffected. |
