| molecularNote | Using transalleleic targeted meiotic recombination, this allele was created in which both the Prnd and Prnp loci are disrupted. The recombined allele lacks ~16 kb encompassing both genes. To generate this allele, Prnptm2.1Cwe mice, which have a loxP site in place of the coding sequence, were crossed with Prndtm1Aag mice, which have a floxed neo in place of the coding exon. Northern blot analysis indicated absence of Prnp and Prnd transcript in brain, spleen and testes of mutants. |
