| molecularNote | This mutation consists of an insertion of a Sleeping Beauty transposable element gene trap construct into the first intron of this gene. The Sleeping Beauty transposon construct includes a Carp beta-actin splice acceptor and exon sequences with stop codons in all three reading frames, followed by the EMCV internal ribosome entry site (IRES) and Clontech's Tet-off cDNA, with an SV40 polyadenylation signal, all between the inverted terminal repeats of the transposon (IR/DRs), This mutation was generated by germline mobilization of chromosomally-resident transposons from concatemer line 4563. As a consequence, this particular transposon insertion is not unique in the carrier strain. Multiple other linked insertions may exist with this insertion, although none of the others have been shown to be in genes at the time of submission. |
