abstractText  The Numbl targeted mutation allele, a loxP site and FRT site flanked targeting vector containing a neomycin resistance gene under the control of the phosphotransferase promoter was utilized. This selection cassette was inserted upstream of exon 1 of the targeted gene, and another loxP site was inserted upstream of exon 4. This construct was electroporated into 129/Sv derived GS1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into recipient blastocysts. The resulting chimeric mice were bred to Flp recombinase expressing transgenic mutant strain to remove the selection cassette. The mice were then bred to mice of unknown genetic backgrounds. Mice that are homozygous for these alleles are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities.
