| description | This is one of 11 lines generated using this transgenic construct. Of the 11, five lines were characterized for their effect on expression of a tetO-lacZ reporter gene in doubly transgenic mice. Such mice, when treated with doxycycline, exhibit 3- or 30-fold elevated levels of beta-galactosidase (b-gal) activity in skin relative to levels in tetO-lacZ singly-transgenic mice. Immunohistochemical staining for b-gal detects expression in skin and hair follicles, specifically in the outer root sheath of hair follicles and, in one line, the basal layer of the epidermis. Lower-level induction occurs in tongue, forestomach and trachea. No significant b-gal is detected in tissues that do not express endogenous keratin 5 or in the absence of doxycycline. (J:65791) |
| molecularNote | This transgene contains the reverse tetracycline-regulated transactivator gene rtTA under control of the bovine keratin 5 promoter. The rtTA coding sequence lies downstream of approximately 5.2 kilobases of DNA from the 5' flanking region of the keratin followed by a beta-globin intron and upstream of an SV40 polyadenylation signal. |
