molecularNote | This transgene comprises a human growth hormone gene whose 5' regulatory elements have been replaced by nucleotides -596 to +21 from the rat liver fatty-acid binding protein gene. Mice of this line (from founder G0#19) have ~800 transgene copies per haploid genome, as measured by densitometric analysis of Southern blots. Expression of human growth hormone mRNA, determined by dot-blot analysis, correlates generally with that of endogenous mouse Fabp1 mRNA in liver and intestine, including a proximal-to-distal declining expression gradient in the small intestine (with somewhat higher hGH expression in the ileum); neither is expressed in several other tissues examined, but hGH, unlike Fabp1, is expressed in cecum, colon and kidneys of transgenic mice. This "ectopic" expression is suppressed by more 5' regulatory sequences present in similar transgenes with a longer Fabp1 promoter (See Tg(Fabp1-GH1)5Bir). |
