| molecularNote | A targeting vector was designed to change the codon within exon 4 that encodes the glutamic acid at position 16 into that of a proline (E16P). A floxed betageo selection cassette was included downstream of exon 5. Mice carrying the targeted mutation in the germline were crossed with cre transgenic mice to remove the selection cassette, leaving behind a single loxP site. Correct targeting was confirmed by genomic PCR. The amino acid substitution enhanced the affinity of the protein for the InlA surface protein expressed by Listeria monocytogenes. |
