| molecularNote | The entire coding region of the locus was replaced by a Gfp reporter gene and a loxP-flanked neomycin resistance cassette. Chimeric mice were crossed with a cre-deleter strain to remove the neo cassette, leaving behind a single loxP site. The mutated allele has the Gfp gene inserted in-frame with the endogenous start and stop codons. Fetal livers from E13.5 homozygous embryos have expression of GFP but no expression of the endogenous gene locus, as determined by immunoblot analysis. GFP expression is visible by fluorescent microscopy in both heterozygote and homozygote E13.5 embryos. |
