molecularNote | The Ple185-EGFP transgene (pEMS1384) was designed with a Ple185 minipromoter (S100beta-C; a 2440 bp promoter segment and 1340 bp basal promoter element, both from the human S100 calcium binding protein B (S100B) promoter region) upstream of a minimal F5 mutant-frt site, an enhanced green fluorescent protein (with mutated TAA->TTA stop), a nuclear localization signal, a second minimal frt site, an SV40 early polyA signal, and a human HPRT complementary sequence (containing exon1, intron1, exon2, and part of intron2). This construct was targeted as a single copy knockin to the Hprtb-m3 mutant locus on the X chromosome. The promoter/regulatory regions of the human S100 calcium binding protein B (S100B) gene directs expression of enhanced green fluorescent protein (EGFP) to neural white matter glia and astrocytes. There is broad EGFP expression throughout the brain in some glial populations, most notably in cerebellar Bergmann glia, as well as fiber tracts throughout the brain. Descending white matter fiber tracts coursing through the corpus callosum, striatum, and into the brainstem are also prominently labeled. The EGFP expression pattern in the brain co-labels with myelinated axons in the cortex, striatum, and brainstem. |
