| molecularNote | An inversely oriented FRT-flanked neomycin cassette replaced 1.7 kb of the 5' sgamma1 switch region that is upstream of the IgG1 constant region and includes sites of DNA modification and a splice donor site. After targeting 129P2/OlaHsd ES cells, the neomycin cassette was removed by transient expression of Flpe recombinase. Inactivation of the IgG1 switch region was confirmed in (C57BL/6 x 129P2/OlaHsd)F1 mice by a lack of IgG1 with the "a" 129 haplotype (i.e. IgG1a). |
