| molecularNote | The gene trap vector (VICTR48) was inserted into intron 6, resulting in truncation of the carboxyl-terminus. The gene trap allele was expected to generate two separate transcripts, the marker fusion transcript (MFT) consisting of a truncated 5' region and the Neo selection marker, and the OmniBank sequence tag (OST) fusion transcript (OFT). RT-qPCR analysis revealed a ~90% reduction in 5' transcripts and nearly undetectable 3' transcripts (OFT RNA) in P14 mutant bladders. Immunoblot analysis using an antibody that recognizes an N-terminal epitope revealed only a faint signal from mutant bladder tissue. |
