| molecularNote | The transgene construct encodes mouse MAS-related GPR, member A1 joined at its C-terminus to green fluorescent protein; expression of this fusion protein is regulated by a "tet minimal promoter" comprising seven copies of the tet operator sequence followed by the human cytomegalovirus minimal immediate early (CMV-IE) promoter. The construct also contains, interposed between the promoter and the coding sequence, exon 1, intron 1, and 10 base pairs of exon 2, including the splice acceptor, from the mouse transthyretin gene; and following the coding sequence, the SV40 early region polyadenylation signal. In mice bearing this transgene and expressing a tissue/cell type-specific tetracycline transactivator (tTA) or reverse transactivator (rtTA), expression of GFP-tagged MRGPRA1 is regulated by the presence or absence of doxycycline. |
