molecularNote | This transgene consists of human RAMP1 cDNA under the control of the CX1 promoter, which is a hybrid of the chicken beta-actin promoter and CMV enhancer. The CX1 vector into which RAMP1 cDNA was inserted also contains an eGFP gene flanked by loxP sites following the promoter (and upstream of RAMP1) and a beta-actin intron and bovine globin polyadenylation signal. CX1 drives ubiquitous expression of GFP, but not RAMP1 due to a translation stop codon and a polyadenylation signal in the GFP cDNA. Not until cre-mediated excision of the GFP-stop sequence at the flanking loxP sites occurs, that RAMP1 is expressed. |
