| molecularNote | Homologous recombination in bacterial cells inserted a construct with cre recombinase fused with the tamoxifen-responsive mutated estrogen receptor sequence, ERT2, into the ATG uptstream starting coding region of the tissue-nonspecific alkaline phosphatase gene (Alpl) in BAC clone RP23-400F8. The correctly targeted BAC was electroporated into fertilized eggs, producing seven founders. These were crossed to R26R reporter mice and three lines (3, 6, and 12) showed efficient cre recombination in progeny. Most subsequent experiments were performed with the line propagated from founder 12 which was more fertile than the other founders. |
