molecularNote | A cDNA encoding human signaling lymphocytic activation molecule family member 1 (also called CD150) was inserted into the ubiquitous expression vector pHMG, which contains the promoter, untranslated first exon and first intron of the mouse 3-hydroxy-3-methylglutaryl-Coenzyme A reductase gene and the SV40 polyadenylation signal. Human SLAMF1 mRNA was detected by reverse transcription-polymerase chain reaction (RT-PCR) in multiple tissues of transgenic mice; the highest expression was observed in brain. Flow cytometry using an anti-SLAMF1 monoclonal antibody followed by FITC-conjugated goat anti-mouse antibody demonstrated the presence of human SLAMF1 on the surfaces of cells from various regions of transgenic mouse brains, including cortex, cerebellum/spinal cord, and hippocampus. |
